dc.description.abstract | La papa es uno de los cultivos más importantes en el mundo y en Canarias, y en este trabajo tratamos de optimizar un protocolo de esterilización y de microtuberización para la variedad ‘Negra yema huevo’ (Solanum chaucha). Esta variedad pertenece a las Papas Antiguas de Canarias que son Denominación de Origen Protegida (DOP). Mediante el cultivo in vitro de las Papas Antiguas de Canarias se puede perpetuar su germoplasma, ya que se está disminuyendo su cultivo, y ello supondría una pérdida incalculable. Para la esterilización del material vegetal se idearon 4 protocolos combinando diferentes concentraciones de lejía (< 5% hipoclorito sódico) y fungicida (16,7 g/L difenoconazol) para intentar eliminar la contaminación procedente de campo. Los protocolos con 20 ml/L de fungicida fueron los más eficaces en la esterilización, con independencia de la concentración de hipoclorito sódico. Una vez esterilizados los explantes de papa, se establecieron in vitro en un medio MS0, estudiándose como la esterilización pudo afectar a su crecimiento. El protocolo con las concentraciones más bajas de lejía (30 %) y fungicida (10 ml/L) resultó ser en el que los explantes tuvieron un mayor crecimiento. Posteriormente se indujo la microtuberización en 12 medios con diferentes concentraciones de hormonas y sacarosa, en condiciones de luz y oscuridad, observándose que los mejores tratamientos eran aquellos que incluían en el medio 2,5 mg/L de benciladenina (BA), 8% de sacarosa y con un fotoperiodo de 16 h luz/8 h oscuridad. El cloruro de clorocolina (CCC) a 0,5 g/L, también resultó óptimo en ciertas condiciones, pero no esencial para el desarrollo de los microtubérculos. | es |
dc.description.abstract | The potato is one of the most important crops in the world and in Canary Islands, and in this work, we try to optimize a sterilization and microtuberization protocol for the ‘Negra yema huevo’ variety (Solanum chaucha). This variety belongs to the Ancient Potatoes of the Canary Islands that are Protected Designation of Origin (PDO). Through the in vitro cultivation of the Ancient Potatoes of the Canary Islands, their germplasm can be perpetuated, since their cultivation is decreasing, and this would represent an incalculable loss. For the sterilization of the plant material, 4 protocols were devised combining different concentrations of lye (<5% sodium hypochlorite) and fungicide (16.7 g/L diphenoconazole) to try to eliminate contamination from the field. The protocols with 20 ml/L fungicide were the most effective in 2 sterilization, regardless of the concentration of sodium hypochlorite. Once the potato explants were sterilized, they were established in vitro in an MS0 medium, studying how sterilization could affect their growth. The protocol with the lowest concentrations of lye (30%) and fungicide (10 ml/L) was the one in which the explants had the highest growth. Subsequently, microtuberization was induced in 12 media with different concentrations of hormones and sucrose, in light and dark conditions, observing that the best treatments were those that included in the medium 2.5 mg/L of benzyladenine (BA), 8% of sucrose and with a photoperiod of 16 h light/8 h darkness.
