Estudio de diferentes medios de cultivo in vitro en la propagación del género Phalaenopsis a partir de protocormos

En este Trabajo Fin de Grado se describe el procedimiento para evaluar la
regeneración de Phalaenopsis y Cymbidium a 17ºC ±1ºC de temperatura
constante. En Phalaenopsis se utilizan cinco medios diferentes, con cinco
repeticiones cada uno y en Cymbidium, se ensaya dos medios con diez
repeticiones cada uno. Para determinar si hay disparidad en el desarrollo de las
plántulas en los diferentes medios se hace mediante un análisis de varianza
(ANOVA p<0,05) y prueba de Tukey, con medidas tomadas de hojas y raíz,
utilizando dos tipos de contenedor. En ambos ensayos se utiliza un volumen de
200 cm3 de medio sólido por recipiente a 17ºC ±1ºC constante con 16 horas de
luz artificial y 8 horas de oscuridad. Se comprueba la aclimatación en las
condiciones externas de cultivo en umbráculo, bajo malla del 80%. A partir de
la primera semana y, hasta la semana final del ensayo, se hizo un seguimiento
del cultivo in vitro. Además, se realizó un seguimiento fotográfico. Entre los
resultados más relevantes se constata que, en los tipos de contenedores y
medios de cultivo evaluados a 17ºC ±1ºC, no se observa ninguna anomalía
fisiológica ni mortalidad. Respecto al medio de cultivo en Phalaenopsis y
Cymbidium, Hyponex, según estadística, fue el medio con mejor desarrollo
vegetativo y radicular. La evolución de las plántulas ex vitro no presentandaños
por aclimatación. This Final Grade Project describes the procedure to evaluate the regeneration
of Phalaenopsis and Cymbidium at 17ºC ± 1ºC of constant temperature. In
Phalaenopsis five different media are used, with five repetitions each, and in
Cymbidium, two media with ten repetitions each are tested. To determine if
there is disparity in the development of the seedlings in the different media, it is
done through an analysis of variance (ANOVA p <0.05) and Tukey's test, with
measurements taken from leaves and roots, using two types of container. In
both tests, a volume of 200 cm3 of solid medium is used per container at a
constant 17ºC ± 1ºC with 16 hours of artificial light and 8 hours of darkness.
Acclimatization is checked in the external conditions of cultivation in shade,
under an 80% mesh. From the first week and until the final week of the test, the
in vitro culture was monitored. In addition, a photographic examination was
carried out. Among the most relevant results, it is found that, in the types of
containers and culture media evaluated at 17ºC ± 1ºC, no physiological
abnormality or mortality was observed. Regarding the culture medium in
Phalaenopsis and Cymbidium, Hyponex, according to statistics, was the
medium with the best vegetative and root development. The evolution of the
seedlings ex vitro does not show damage by acclimatization.
