They are nanocarriers that combine the positive attributes of both liposomes and polymeric nanoparticles and they stand out for their ability to modify thebiodistribution profile, solubility and bioavailability of the encapsulated active ingredient.Its preparation by conventional methods presents several obstacles such as batch to batch reproducibility and variations in the physicochemical properties. Due to the sensitivity of these production techniques, it is very important to understand what factors influence the process and, therefore, the formation of nanoparticles. Among them we canhighlight the nature of pegylated lipids, the type of polymer and the lipid/polymer ratio, which can modify the size, the polydispersity index and the zeta potential.In this preliminary study, we will proceed to the elaboration of hybrid nanoparticles by modified nanoprecipitation and the optimization of different factors related with the formulation. In this way, it will be possible to establish a formulation with suitable characteristics for the encapsulation of a GapmeR.The results show that the mean particle size increases linearly with polymer concentration, while the polydispersity index decreases potentially. Likewise, the decrease in the lipid/polymer ratio up to 15% does not produce substantial differences inthe physicochemical properties with respect to formulations with a higher ratio. Finally, only a slight decrease in the zeta potential was observed for nanoparticles made with DOPE-MPEG2000 compared to those made using DSPE-MPEG2000.
