PTENmutations alter PIP3 mediated-signaling, affecting cellular processes such as apoptosis,metabolism or cell growth. Yeast PIP3 levels are undetectable; however, PTEN is aubiquitous gene in fungi. Our group has identified a PTEN orthologue in yeastHansenula polymorpha, named HpTep1, through a screening method to identifymutants affected in nitrate assimilation. Previous works on Tep1 shows that TEP1deleted strain rise PIP3 levels and is able to downregulate gene expression levels ofgenes involved in nitrate assimilation. This strain has also lower nitrate reductaseactivity. This work is on focus to study if Tep1 is altering the Ynt1 high affinity nitratetransporter intracellular trafficking in H. polymorpha. Our results show that Δtep1 strainwas sensitive to toxic amino acid analogues and showed greater nitrite excretion levels,thus Ynt1 could be accumulated on the plasma membrane. Analysis of Ynt1 levels byWestern-blotting confirms this hypothesis, suggesting that Tep1 could have animportant role in the intracellular trafficking regulation of transporters in yeast.
