Evaluación de ensayos de citotoxicidad frente a Redes Metal-Orgánicas
Abstract
Metal-organic frameworks (MOFs) are novel crystalline materials that are gaining
a lot of attention in the environmental monitoring field. However, very little is known
about the possible health or environmental risks that they may provoke. In the present
study, six MOFs, specifically UiO-64, UiO-66, CIM-80, CIM-81, CIM-82 and CIM-84,
were synthesized and characterized. Two of them, UiO-64 and UiO-66 are MOFs widely
used; the other MOFs, with the abbreviations CIM (Canary Islands Materials), have been
characterized in the University of La Laguna, and CIM-82 and CIM-84 are still in the
phase of solving their crystalline structure. The cytotoxicity of these molecules was
evaluated in vitro with murine macrophages J774.A1 cell line, using the alamarBlue®
method to obtain the lethal concentration that reduces 50% of these cells (CC50).
Furthermore, it was evaluated their cytotoxicity in vivo using amphipod (Gammaropis
atlantica). CIM-81 and CIM-82 were toxic in macrophages (CC50 of 0,98 ± 0,08 mg/mL
and 0,88 ± 0,01 mg/mL, respectively) and in amphipods (all the individuals were dead).
Therefore, there is a strong correlation between their in vivo and in vitro toxicity results.
Two MOFs (UiO-64 and UiO-66) were more toxic to amphipods that to macrophages.
CIM-80 and CIM-84 were not toxic. Las redes metal-orgánicas (MOFs) son nuevos materiales cristalinos que están
ganando mucha atención en el campo del monitoreo ambiental. Sin embargo, se sabe muy
poco acerca de los posibles riesgos para la salud o el medio ambiente que pueden
provocar. En el presente estudio, se sintetizaron y caracterizaron seis MOFs,
específicamente UiO-64, UiO-66, CIM-80, CIM-81, CIM-82 y CIM-84. Dos de ellos,
UiO-64 y UiO-66 son MOFs ampliamente utilizados; los otros MOFs, con las
abreviaturas CIM (del inglés: Canary Islands Materials), se han caracterizado en la
Universidad de La Laguna, y CIM-82 y CIM-84 aún se encuentran en la fase de
resolución de su estructura cristalina. La citotoxicidad de estas moléculas se evaluó in
vitro con la línea celular J774.A1 de macrófagos murinos, utilizando el método
alamarBlue® para obtener la concentración letal que reduce el 50% de estas células
(CC50). Además, se evaluó su citotoxicidad in vivo utilizando anfípodos (Gammaropis
atlantica). CIM-81 y CIM-82 fueron tóxicos en macrófagos (CC50 de 0,98 ± 0,08 mg/mL
y 0,88 ± 0,01 mg/mL, respectivamente) y en anfípodos (todos los individuos murieron).
Por lo tanto, existe una fuerte correlación entre los resultados de toxicidad in vivo e in
vitro. Dos MOFs (UiO-64 y UiO-66) fueron más tóxicos para los anfípodos que para los
macrófagos. CIM-80 y CIM-84 no fueron tóxicos.
